Haemoproteus in passerines: Prevalence and infekction dynamics

The genera Haemoproteus and Plasmodium are widespread genera of blood parasites from phylum Apicomplexa. Both genera have dixenous life cycle and their definitive hosts belong to the order Diptera. Both parasites can be found in birds where asexual division takes place. The infection with genera Haemoproteus and Plasmodium is usually asymptomatic; during long lasting chronic phase relapses of the infection can occur. We gained 1 092 blood samples from 29 trapped species of passerines in Milovice forest during seasons 2017-2019. Haemosporidians were detected in 48 % of samples. The prevalence of genus Plasmodium was 16 %, the prevalence of genus Haemoproteus was 22 % and 10 % we could not specify the parasite. We have found six new lineages of genus Haemoproteus, provisionally named Haemoproteus lineages coccoc_1, coccoc_2, coccoc_3, coccoc_4, embcit, fricoe which total prevalence was 14 %. Lineage Haemoproteus sp. coccoc_1 was the most noticed one and its prevalence was 12 %. Plasmodium relictum clone Peng14-121Br2AF and isolate Cc_P1 was the second most prevalent (13 %). The role of biting midges in avian blood parasites transmission

Biting midges of the genus Culicoides (Diptera: Ceratopogonidae) are hematophagous in- sects mainly important as vectors of viral diseases of farm animals. They have shifted to the forefront of scientific interest in the Czech Republic primarily in relation to the Bluetongue Virus spread since 2007. However, they have been often ignored as vectors of parasitic infec- tions, especially if these are not pathogens of economic importance. In birds, which are the focus of this bachelor's thesis, biting midges are the most significant vector mainly for the genus Haemoproteus, they transmit at least 24 species of these protozoa. On the other hand, they are proven vectors for only one species of the genus Leucocytozoon - L. caulleryi. Trans- mission has been proven for three species of the genus Trypanosoma - T. avium, T. bennetti and T. everetti. Biting midges also transmit helminths of the family Onchocercidae, they play a role as vectors for the genera Eufilaria, Chandlerella and Splendidofilaria. Avian haemosporidia: Prevalence, host specificity, vectors

Haemosporidia are a group of protozoan parasites that shifts between vertebrates and insect vectors during a complicated life cycle. In this thesis I have focused on two of the genera infecting birds, Plasmodium and Haemoproteus (sometimes together referred to as avian malaria) in terms of their host specificity and prevalence in avian hosts and vectors. These globally distributed genera have different vectors, and although they both attack bird hosts, they have different host specificities. Avian malaria parasites and their vectors

Parasites causing avian malaria belong to the group Haemosporida, which represents a monophyletic group of dixenic protists within Apicomplexa. Their asexual reproduction takes place in a vertebrate intermediate host, and the formation of gametes and sporogony occur in blood-sucking dipteran insects, which are the definitive hosts of these parasites. Three main genera (​Plasmodium​, Haemoproteus ​and ​Leucocytozoon​) are found mostly in their avian hosts. We focused on the Haemosporida of wild birds and their transmission by insect vectors in natural populations, which had previously been a neglected area. Our results were obtained both by traditional methods (investigation of infections by microscopy of blood smears) and mainly by molecular methods (e.g. nested PCR) centered around work with unique haplotypes of the haemosporid lineages. The aim of our work was to determine the range of possible insect vectors of avian haemosporidians in the territory of the Czech Republic, taking into account the specificity of the parasites within these vectors, and to describe the diversity of haemosporidians in the populations of their bird intermediate hosts. The genus Haemoproteus is worldwide distributed parasitic protist, which belongs to the phylum Apicomplexa. This blood parasite has dixenous life cycle and he is occurs in the bird's blood and in invertebrate vectors. Genus Haemoproteus is transmitted by blood-sucking vectors, biting midges (Ceratopogonidae) and hippoboscid flies (Hippoboscidae). Vectors are final hosts of the genus Haemoproteus and the sexual process takes place in these vectors. Birds are intermediate hosts, in which representatives of the genus Haemoproteus spend most of their lives. In this bachelor thesis I focused on the influence of the genus Haemoproteus on birds and I focus on the time, when the bird gets infected by Haemoproteus in his life. Avian malaria in the Swallow

Long-distance migratory birds can encounter a wide range of parasites. Various populations of birds within one species use different migration routes and can also winter in different places. It can be supposed that birds which use different migration routes should be infected with different parasites. To study the relationship between the migration and the distribution of parasites we chose the worldwide species barn swallow (Hirundo rustica) and the avian malaria parasites. Swallows migrate long distances in different migrating routes. Some populations of swallows do not migrate, they are resident. This is the reason why swallow is a good model species for finding the answers to questions whether the populations using different migration routes are infected with different parasites or not and whether or not the diversity of parasites is wider in populations which migrate long distances in comparison with the resident populations. The malaria lineages of the genus Plasmodium and Haemoproteus were detected using nested PCR and sequencing. 1242 samples from 8 different localities from the USA, Europe and Asia were tested. Can avian malaria affect the reproductive success of the host?

Malarial deseases caused by intracellular parasites of birds is very common. Even if infection doesn't lead to the death of infected individuals, they may suffer alternations of different levels of fitness, which may also consequently harm their reproductive success rates. Infected individuals, due to their poor physical condition, may lay lower quality eggs, breed less viable offspring, have reduced frequency of feeding their chicks, etc. While it may seem that malarial parasites undoubtedly negatively affect their hosts's reproduction, there are numerous studies which do no support such prediction. These differences in individual studies may be due to various testing methods used in the indicated studies. Because of various testing methods, there also may appear other possible problems which can more or less affect the detection rate of malaria parasites.
